8 compelling reasons to consider our icIEF-MS: 1) Regulatory compliance in the evolving field of protein therapeutics can be difficult to navigate. Stay up-to-date with the most recent tools for charge variant characterization! 2) Do you spend weeks (or months) on method development for LC-MS? With icIEF-MS it can take as little as 1 day. 3) Does an unexpected peak appear on your icIEF profile during QC monitoring? Avoid costly delays in manufacturing and investigate the issue with icIEF-MS. The method transfer from your current icIEF instrument is seamless. 4) Do you need more in-depth charge variant characterization? Our instrument also has an offline fractionation mode to collect larger quantities (~50-200ug) of each icIEF peak for peptide mapping, amino acid sequencing, and more. Once again, the method transfer from your current icIEF instrument is seamless. 5) Do you want to couple directly to your existing MS instrument? We are compatible with a variety of commercial instruments, including the Orbitrap which is unmatched in terms of resolution, sensitivity, and accuracy. 6) Are you concerned about reproducibility which has traditionally been an issue for CE-MS? We have addressed many of these technical challenges with our proprietary reagents and patented pressure mobilization technique. 7) Our icIEF-MS cartridges offer excellent durability and cost savings (~100 runs) thanks to our unique manufacturing protocols. 8) We continue to develop new features and methods based on our customers’ needs. The most recent example is our nanospray interface which has simplified the connectivity and improved sensitivity. Agitation of therapeutic protein solutions during manufacturing, shipping and handling is one of the major initiators for protein aggregation and particle formation during the life history of a protein drug. Adsorption of protein molecules to liquid-air interfaces leads to the formation of highly concentrated protein surface films. The rupture of these protein films due to various mechanical processes can then result in the appearance of protein aggregates and particles in the bulk solution phase. One technique to stabilize proteins against stress induced by liquid-air interfaces is the use of non-ionic surfactants. This study compares various molecules for their capabilities to reduced agitation-induced protein aggregation and particle formation, and investigates their underlying protein stabilizing mechanisms. Learn more about it in our recent whitepaper published by Michelle P. 🧪Interesting developments of Protein A Affinity Resins! The latest article discusses cutting-edge techniques and technologies to enhance the binding capacity of affinity resins, boosting their performance in downstream purification. Among the methods explored is site-directed immobilization via a terminal cysteine residue of the recombinantly expressed Protein A, known for its efficiency in increasing binding capacity. Additionally, oligomerization of the Protein A motifs has shown promising results by improving steric accessibility and thus enhancing binding capacity. In the article, you'll find a comprehensive review of existing spacer technologies in affinity chromatography resins, with a focus on the effective approach of oligomerizing the particular Protein A motifs. This innovative technique has demonstrated remarkable potential, with dynamic binding capacities reaching up to twice the values obtained with traditional tetrameric ligands. However, high protein binding capacity isn't the only important criterion for affinity resins; there are also protein stability, high recovery rates, host cell protein removal, and low protein aggregation. The research also delves into the impact of buffer concentration on binding capacity, revealing exciting insights on achieving up to 130 mg/ml with the hexamer ligand. Equilibrium isotherms further highlight the promising potential of this technology. Check out the full article for more details and stay ahead in the world of biopharmaceutical purification! 13 FAQs about Peptide Synthesis, Click Like & Repost Please~ How Long and How Much Purity is Suitable for Immunological Peptide? Generally about 10-15 amino acids, of course, longer immune effect is better, but the cost of synthesis will also increase. MAP peptide is expected to be longer than 15aa, the effect is better. In addition, the immune effect of peptides below 10aa was poor. What is the Peptide Form? How to Preserve and Dissolve? The peptides we provide are in powder form, generally gray and white, the composition is different, the color of the peptide powder is different, the peptides generally need to be kept away from light for a long time, and should be stored at -20 degrees, and can be stored at 4 degrees in the short term. Transport at room temperature if possible for a short time. How to Dissolve Peptides? Dissolving peptides is a very complex matter, and it is generally difficult to determine the right solvent all at once. Usually a little test is taken first, do not dissolve the joint before determining the appropriate solvent. Omizzur Biotech generally tests several dissolution conditions after peptide synthesis and marks them on the HPLC. Intramodule Pressure Profiles and Protein Accumulation During Tangential Flow Filtration While TFF has been used to remove casein from milk for cheese production for over 35 years, and in pharmaceutical manufacture of biotherapeutic proteins for 20 years, the rapid decline in filtration rate (i.e., flux) at high protein concentrations is a limitation that still needs to be addressed. This is particularly important for mAbs, many of which are 140–160 kDa immunoglobulin G (IgG) type proteins recovered at concentrations of 200 mg/mL or higher. This work reports the direct measurement of local transmembrane pressure drops and off-line confocal imaging of protein accumulation in stagnant regions on the surface of a 30 kDa regenerated cellulose membrane in a flat-sheet configuration widely used in manufacture of biotherapeutic proteins. https://lnkd.in/gu-JF9yi #aspenalert #biotech #bioprocess
